Van Fleuten first competed in the London Olympics in 2012, where she competed in the line race and finished 14th, while her compatriot Marianne Vos won the race.

Van Vleuten fell down a hill from Vista Chinese on August 7, 2016, while leading the line race of the 2016 Summer Olympics, after failing to brake in a steep corner 12 kilometers from the target. She was taken to the hospital with three lower back fractures and a serious concussion as a result of the accident.
